Necessary Evil: The CIP in Process Scale HPLC

Insulin, insulin analogues and related diabetes treatment peptide based APIs are the biggest application for silica based HPLC in process scale.

These molecules are prone to self-aggregation and fibrillation rendering the big silica packed columns useless in intolerably short time.
To remove the fibrillated peptide layer from the column unspeakably the silica is treated usually with pH 13 NaOH solution. This latter damaging the silica based stationary phase and again shortening the useful time of the column. Breaking out of this predicament is much needed, but before that we need scientific tools to study the actual situation of the “dirtiness” of the column and the damage suffered by the silica.

What is Industrial-scale CPC and How Does it Outperforms Traditional Liquid Chromatography?

Centrifugal Partition Chromatography (CPC) is one of the technical realization of liquid-liquid chromatographic, where the liquid stationary phase is immobilized by a strong centrifugal force, coming from a circular motion. A series connected network of extraction cells, which operates as elemental contionous extractors, guarantees the efficiency by the cascade. CPC instruments vary on scale from 50 ml to 25 liter, however all advantages are realized on bigger scale use in industry as it provides a more cost effective and more efficient separation to preparative HPLC. Up to date there were no real available instruments, those could be used for API purification in real situations.

By industrial size we mean process chromatography, the same scale that can be achieved by conventional HPLC DAC columns, with diameter of 150-1200 mm, and flow rate of 1 to 20 liters / minute. So far tt was impossible for the CPC to get a role in API purification without this ability.

In this presentation, we would like to introduce the basics of CPC, and highlight the development steps that provided the pharmaceutical industry the first real industrial CPC, the RotaChrom iCPC. Starting from theoretical basics of the flow simulations. At last, we would like to present applications of our new technology from different examples of API purification in the pharmaceutical industry, compared to preparative HPLC and other techniques, proving that prep HPLC is the past for API purification.

Latest Stationary Phase Choices for Preparative Chromatography

The seminar will focus on the latest developments of Kromasil materials including EternityXT, a stationary phase with organo silane merged into the base silica matrix. EternityXT , with its high chemical and mechanical stability, as well as its scalability, makes it possible for users to work under tough pH conditions and higher temperatures across the various stages of product development including discovery, pilot plant and manufacturing.
